A Berry Bowl of Martian Spherules

How were these unusual Martian spherules created? Thousands of unusual gray spherules made of iron and rock and dubbed blueberries were found embedded in and surrounding rocks near the landing site of the robot Opportunity rover on Mars in 2004. To help investigate their origin, Opportunity found a surface dubbed the Berry Bowl with an indentation that was rich in the Martian orbs. The Berry Bowl is pictured here, imaged during rover's 48th Martian day. The average size of a Martian blueberry rock is only about 4 millimeters across. By analyzing a circular patch in the rock surface to the left of the densest patch of spherules, Opportunity obtained data showing that the underlying rock has a much different composition than the hematite rich blueberries. This information contributes to the growing consensus that these small, strange, gray orbs were slowly deposited from a bath of dirty water.

President Donald Trump has claimed that Tehran’s nuclear facilities were “completely and totally obliterated”

Iran’s Fordow nuclear site did not suffer any serious damage in the attack launched by the US, Fars news agency reported on Sunday, citing MP Mohammad Manan Raisi from Qom Province where the facility is located.

US President Donald Trump has claimed that the Fordow, Natanz and Esfahan nuclear sites were “completely and totally obliterated” in the American strikes overnight.

Raisi told the agency that “contrary to the claims of the lying US president, the Fordow nuclear facility has not been seriously damaged, and most of what was damaged was only on the ground level and can be restored.”

The lawmaker described the effect of the American bombing as “superficial,” stating further that there were no fatalities and no radiation leaks at the site afterward.

“This US aggression means that America is directly entering the war, and now it is Iran that determines how and in what way to respond to this obvious American stupidity,” Manan Raisi

Reuters reported earlier, citing an unnamed high-ranking Iranian official, thatmost of the enriched uranium had been moved from Fordow to an undisclosed location before the US strike.

Iran’s Center for the National Nuclear Safety System confirmed the attack on the country’s nuclear facilities, but said that emergency inspections at the affected sites had found no signs of radioactive contamination or leaks. (So they are admitting these were nuclear materials)

The International Atomic Energy Agency also said later that no increase in radiation levels had been reported at Fordow, Natanz, and Esfahan.

The Islamic Revolutionary Guard Corps (IRGC) issued a statement on Sunday accusing the US of launching an “illegal military attack on the peaceful nuclear facilities” in coordination with Israel, and in violation of the UN Charter and international law.

The Americans and Israelis “neither have the power of initiative nor the ability to escape heavy responses” by Iran, the IRGC said, adding that it has already identified the locations where the planes that took part in the attack are stationed.

This was the ninth test flight for the rocket, and the third catastrophic failure in a row, just this year.

Is this what we should expect from the very ship some are counting on to take humans further than we’ve ever been in the solar system? Or does this failure point to deeper concerns within the broader program?

The Starship program from Elon Musk’s space technology company, SpaceX, has been in development for more than a decade now and has undergone many iterations in its overall design and goals.

The Starship concept is based upon the SpaceX Raptor engines to be used in a multistage system.

In a multistage rocket system, there are often two or three separate blocks with their own engine and fuel reserves. These are particularly important for leaving Earth’s orbit and travelling to the Moon, Mars and beyond.

With Starship, the key factor is the ability to land and reuse vast amounts of the rocket stages again and again. The company’s Falcon 9 vehicles, which used this model, were fantastically successful.

Initial tests of Starship began in 2018 with two low-altitude flights showing early success. Subsequent flights have faced numerous challenges with now four complete failures, two partial failures and three successes overall.

Just two days ago, during the latest failed attempt, I watched alongside more than 200 other space industry experts at the Australian Space Summit in Sydney.

Broadcast live on a giant screen, the launch generated an excited buzz – which soon turned to reserved murmurs.

Of course, designing and launching rockets is hard, and failures are to be expected. However, a third catastrophic failure within six months demands a pause for reflection.

On this particular test flight, as Starship positioned itself for atmospheric re-entry, one of its 13 engines failed to ignite.

Shortly after, a booster appeared to explode, leading to a complete loss of control. The rocket ultimately broke apart over the Indian Ocean, which tonnes of debris will now call home.

Polluting Earth in pursuit of space

We don’t know the exact financial cost of each test flight. But Musk has previously said it is about US$50–100 million.

The exact environmental cost of the Starship program – and its repeated failures – is even harder to quantify.

For example, a failed test flight in 2023 left the town of Port Isabel, Texas, which is located beside the launch site, shaking and covered in a thick cloud of dirt.

Debris from the exploded rocket smashed cars. Residents told the New York Times they were terrified. They also had to clean up the mess from the flight.

Then, in September 2024, SpaceX was fined by the US Environmental Protection Agency and the Texas Commission on Environmental Quality for 14 separate incidents since 2022 where the launch facilities discharged polluted water into Texas waterways.

Musk denied these claims.

SpaceX is targeting Sunday, June 22 for a Falcon 9 launch of the Transporter-14 mission to low-Earth orbit from Space Launch Complex 4E (SLC-4E) at Vandenberg Space Force Base in California.

The 57-minute launch window opens at 2:18 p.m. PT. If needed, there is a backup opportunity on Monday, June 23 at the same time.

However, a new idea challenges this theory, suggesting that time, specifically in three dimensions, is the true fabric of the universe, with space emerging as a secondary effect..

This strange theory is proposed by Dr. Gunther Kletetschka, a physicist at the University of Alaska Fairbanks.

His research challenges one of the most deeply held ideas in modern physics: that space and time form the joint foundation of the universe.

Kletetschka suggests that time is not a single, forward-flowing line but a multidimensional framework with three independent directions.

He claims that “viewing time as three-dimensional can naturally resolve multiple physics puzzles through a single coherent mathematical framework.”

A framework that satisfies real-world values

To understand Kletetschka’s proposal, imagine replacing the traditional spacetime framework with a six-dimensional setup: three axes for time and three for space. However, unlike conventional physics, this new theory treats time as the core structure.

Space is no longer fundamental; it’s more like a consequence of how time behaves at different scales and orientations. Kletetschka arrived at this idea by re-examining the role of time in physical equations.

He began with the thought that if space can have three dimensions, perhaps time can, too.

Using this assumption, he developed a mathematical model in which particles and forces interact within a three-dimensional temporal framework.

The important insight is that when this model is applied to fundamental particles, it can reproduce some of their known properties, particularly their masses.

For instance, the Standard Model of particle physics includes detailed data on particles like electrons, muons, and quarks, but it doesn’t clearly explain why these particles have the masses they do.

Kletetschka’s framework generates values that closely match the observed masses of these particles. This clearly shows that he’s not just describing an abstract idea; he’s using it to explain real-world measurements.

“The three-dimensional time framework represents a significant advance in fundamental physics, providing a unified description of nature while making testable predictions,” he said.

“Its mathematical consistency and explanatory power, combined with specific experimental signatures spanning multiple scales and phenomena, suggest it deserves consideration as a candidate theory of quantum gravity and unification,” Kletetschka added.

Multiple dimensions of time, not a new idea

While some theoretical physicists have previously proposed the idea of extra time dimensions, those earlier models largely remained abstract and mathematical, with little connection to measurable physical quantities.

They also faced major challenges; for example, in those models, having more than one dimension of time often led to logical contradictions, such as the possibility of an effect occurring before its cause.

Kletetschka’s theory avoids this problem by designing a mathematical structure that keeps the flow of time ordered across all three time dimensions. In his model, events still unfold in a consistent sequence, even within a more complex temporal framework.

What makes his work stand out is that it is not just a theory but makes concrete predictions.

His framework accurately reproduces the known masses of fundamental particles like electrons, muons, and quarks, which means it connects directly with experimental data in a way previous models did not.

“Earlier 3D time proposals were primarily mathematical constructs without these concrete experimental connections.

My work transforms the concept from an interesting mathematical possibility into a physically testable theory with multiple independent verification channels,” Kletetschka said.

The next steps could involve refining the mathematical framework and identifying specific experiments, especially in particle physics or cosmology, that could test the theory.

If the assumption remains valid in further research, it might offer a new framework for integrating quantum mechanics with gravity, something that physicists have been chasing for generations.

An explosion has occurred at Mar Elias Church in the Syrian capital of Damascus, multiple media outlets reported on Sunday. The Orthodox Christian church was reportedly struck by a suicide bomber during mass.

At least ten people were killed in the attack and 20 others wounded, reports suggested, citing a preliminary evaluation.

Graphic footage circulating online shows the church interior sustained considerable damage, with mangled bodies and large puddles of blood inside.

The blast was followed by gunshots, eyewitnesses told the UAE-based English-language newspaper The National. The origin of the weapons fire was not immediately clear.

Before the deployment of six U.S. Air Force B-2 stealth bombers—each armed with a 30,000-pound GBU-57A/B Massive Ordnance Penetrator—a satellite intelligence report from Maxar Technologies identified "unusual" activity near the entrance of Iran's Fordow uranium enrichment facility.

The observed movement on Thursday and Friday showed "unusual truck and vehicular activity" near the entrance of the underground Fordow complex south of Tehran. This may indicate foreknowledge or heightened alert status at Iran's high-value nuclear sites.

Newsweek cited Maxar's report and provided more color on the unusual movements:

Pictures captured on Thursday and Friday showed "unusual truck and vehicular activity" close to the entrance of the underground Fordow complex south of Tehran, satellite imagery giant Maxar said late on Saturday U.S. time.

A total of 16 cargo trucks were spotted on the access road leading up to the Fordow tunnel entrance on Thursday, but most of the trucks had relocated to one kilometer (0.6 miles) northwest of the access road by the following day, Maxar said.

New trucks and multiple bulldozers had appeared close to the main entrance by Friday, with one truck very close to the main tunnel entrance, the satellite imagery provider said.

Good morning from Camp Justice at Guantanamo Bay.

Some of the people who came off yesterday's war court shuttle from Washington D.C. are meeting with the detainees. Others are getting ready for a brief, mostly ministerial session in the Bali bombing case.

The two courtrooms are mostly dark this summer while the parties await decisions that will determine next steps in the 9/11 case- and while defense lawyers prepare elsewhere for the USS Cole case trial, now scheduled for next year.

Meantime, we expect the defendant in the Bali bombing case to be brought to court tomorrow. The agenda includes a legal motion or two and the judge's continuing efforts to sort out which lawyers are still actively serving on the defense team.

Iran announced the first-ever launch of its multi-warhead Kheibar Shekan ballistic missile during the twentieth wave of Operation True Promise III in the early hours of Sunday.

In a statement, the public relations department of the Islamic Revolution Guards Corps (IRGC) said that this phase of the retaliatory operation marked the debut of the third-generation Kheibar Shekan ballistic missile, successfully striking its intended targets.

The twentieth wave came just hours after the United States claimed to have struck three Iranian nuclear facilities in Fordow, Natanz, and Isfahan.

According to the IRGC, a total of 40 solid- and liquid-fueled missiles were launched at strategic targets across the occupied Palestinian territories on Sunday.

“In this operation, for the first time, the IRGC Aerospace Force deployed the third-generation Kheibar Shekan multi-warhead ballistic missile, employing new and surprising tactics to achieve greater precision, destructive power, and effectiveness,” the statement said.

Main targets included Ben Gurion Airport in Tel Aviv, the regime’s biological research center, and alternative command-and-control sites.

The missiles employed maneuverable warheads capable of adjusting their trajectory during descent, maintaining guidance until impact, and delivering multiple high-explosive, highly destructive payloads.

Sirens reportedly went off only after the missiles scored direct hits, plunging the occupied territories into chaos, as witnessed in several previous waves of Operation True Promise III.

“We declare that the core capabilities of the Armed Forces of the Islamic Republic of Iran have yet to be activated in this sacred defense,” the IRGC statement concluded.

Iranian armed forces have carried out 20 phases of True Promise III so far, targeting strategic and sensitive military and intelligence sites in the occupied territories.

These operations are in response to the continued Israeli aggression that has claimed the lives of more than 400 Iranians since June 13, including military commanders, scientists, university professors, athletes, school students etc.

First up: French President Emmanuel Macron, who took to social media to declare that “peace and security for all in the region must remain our guiding principle.” Aww, how sweet. Calling for peace while holding your buddy’s coat as he storms into the bar to punch someone on the face. So after Israel unilaterally launched its hundred-target missile tantrum across Tehran – complete with residential hits and assassinations – maybe some strong words against that kind of thing are in order?

“France has repeatedly condemned Iran’s ongoing nuclear program and has taken all appropriate diplomatic measures in response. In this context, France reaffirms Israel’s right to defend itself and ensure its security,” Macron posted as the conflict kicked off.

Classic French parenting: ignore the kid setting the house on fire and scold the one who looked at him funny.

Next up, Germany. Surely the new chancellor, Friedrich Merz, can inject some sober logic into the conversation about who just broke the fragile regional peace everyone’s pretending to care about.

“Iran has subsequently threatened to accelerate uranium enrichment again. This nuclear programme violates the provisions of the Nuclear Non-Proliferation Treaty and poses a serious threat to the entire region, especially to the State of Israel. We reaffirm that Israel has the right to defend its existence and the security of its citizens. We call on both sides to refrain from steps that could lead to further escalation and destabilise the entire region,” wrote Merz.

Oh, so now that Israel has attacked Iran, it’s time for restraint? Not before. Not during. After. Like a guy who throws the first punch in a bar fight and then shouts, “Hey! Let’s all calm down!”

Both Macron and Merz say that Iran brought this on itself by enriching uranium. That’s like this: you’ve got a neighbor who lifts weights in his home gym. You see him through the window with a squat rack, bench press, treadmill – getting jacked. And you’re like, “Wow, he’s getting so ripped I’m afraid he might beat me up someday. So I better go over there now and beat him up while I still can.”

That’s basically what Israel did with its “preemptive” strike. And Macron and Merz are cheering it on like, “Totally! That guy was getting too buff. Definitely deserved a missile to the face.”

Then Merz added from the G7 summit in Canada: “This is the dirty work that Israel is doing for all of us. We are also victims of this regime.”

Oh, sorry – didn’t realize the International Atomic Energy Agency (IAEA) had hired Israel as a global bouncer. And remind us, Friedrich: where exactly did Iran touch Germany on the map? Because Europe is far more likely to be “victimized” by waves of immigration facilitated by its own lax policies, and sparked by its own support for regime change wars – like this one is shaping up to be – than by any distant centrifuge.

European Commission President Ursula von der Leyen, the EU’s de facto queen, is now cosplaying as global playground monitor, sending all the boys to their corners.

“Europe urges all parties to exercise maximum restraint, de-escalate immediately and refrain from retaliation. A diplomatic resolution is now more urgent than ever, for the sake of the region’s stability and global security,” she wrote.

Maximum restraint? His remarks came hours after releasing two pre-recorded statements in Hebrew and English responding to a U.S. strike on three Iranian nuclear sites — Fordow, Natanz and Isfahan. Opening the press conference, Netanyahu expressed condolences to those killed and wounded in Iranian missile attacks on Israeli cities. “Our hearts are with the victims of Iran’s heinous assault,” he said, referencing the heavy toll paid in Operation Rising Lion, the name for Israel’s military response. “We embrace the families of the fallen and pray for the full recovery of the wounded.”

Netanyahu reaffirmed his government’s commitment to neutralizing Iran’s nuclear and missile capabilities, defeating Hamas and returning Israeli hostages. “This regime seeks our destruction. That is why we launched this operation — to remove the two concrete threats to our existence,” he said. “We are advancing step by step toward achieving our objectives, and we are very close to completing them.”

Asked whether the operation included plans to target Iran’s Supreme Leader Ali Khamenei, Netanyahu declined to elaborate. “This is not the forum to detail our goals. I believe in fewer words and more action,” he said.

In his Hebrew statement earlier in the day, Netanyahu emphasized that Israel had fulfilled its early pledge to destroy Iran’s nuclear infrastructure. “From the beginning of this operation, I promised that Iran’s nuclear sites would be destroyed, one way or another. That promise has been kept,” he said.

The U.S. strike on three Iranian nuclear facilities overnight on June 21–22 followed a highly intricate plan that entailed more than 125 U.S. aircraft and warships and layers of deception, Defense Secretary Pete Hegseth said during a Pentagon news briefing.

Hegseth said preparation for the mission—dubbed Operation Midnight Hammer—took place over the course of weeks and months, “so that we could be ready when the president of the United States called.”

The U.S. strikes were made a week after Israel launched a series of surprise airstrikes across Iran, aimed at degrading the country’s nuclear programs and military capabilities.

Joining the conflict that Israel initiated, U.S. military planners set Iran’s Fordow uranium enrichment facility as their primary target. With the Fordow facility situated hundreds of feet underground in a mountainous region of Iran, U.S. Air Force B-2 Spirit stealth bombers carrying 30,000-pound bunker-busting bombs, called GBU-57A/B Massive Ordnance Penetrators, offered one of the best options to destroy the facility.

Boeing has been awarded a $70 million contract for the procurement of GBU-57 Massive Ordnance Penetrator precision-guided “bunker buster” bombs for the U.S. Air Force, a U.S. Department of Defense press release said.

Work under the indefinite-delivery/indefinite-quantity sole source acquisition contract (FA8681-19-D-0008) issued by the U.S. Air Force Life Cycle Management Center is expected to be complete by December 31, 2022, the Monday, September 30 release said.

The GBU-57 Massive Ordnance Penetrator (MOP) is a 30,000-pound (14,000 kg) class GPS-guided “bunker buster” bomb employed only by the iconic B-2 Spirit stealth bomber.

The 20-feet (6m) long weapon is designed to attack deeply-buried and hardened bunkers and tunnels. It carries 5,300 pounds of explosive cased in a special high‑performance steel alloy. The Air Force says it can penetrate up to 200 feet (60m) of concrete before exploding.

Monday’s contract announcement marks a significant dollar increase over previous Massive Ordnance Penetrator orders. The last GBU-57 procurement contract issued was for just under $21 million in February 2018.

Neither Monday’s nor the 2018 contract specified the number of bombs to be procured, but in 2011 the U.S. Air Force ordered eight of the weapons plus supporting equipment for $28 million.

In April, Boeing was awarded a $21.6 million GBU-57 Massive Ordnance Penetrator sustainment contract expected to be completed by July 18, 2023.

The GBU-57 is reportedly the world’s largest in-service non-nuclear bomb, significantly heavier than two other well-known massive bombs – America’s 21,000-lb GBU-43/B “Mother of All Bombs” or MOAB that was dropped in Afghanistan in 2017, and Russia’s 22,000-lb ATBIP “Father of All Bombs” rumoured to have been used in Syria the same year. Both the MOAB and FOAB are thermobaric bombs, designed for huge explosive power rather than penetration.

Elon Musk says AI models are trained on "too much garbage."

He said he plans to use Grok to "rewrite the entire corpus of human knowledge."

And then he said he would retrain the latest model of Grok on that revision.

When you're Elon Musk, you don't have to rely on centuries of prevailing human understanding — you can create your own.

"We will use Grok 3.5 (maybe we should call it 4), which has advanced reasoning, to rewrite the entire corpus of human knowledge, adding missing information and deleting errors," Musk wrote on X on Friday night.

Then, he said he would retrain Grok's latest model on that new base of knowledge to be free of proverbial waste. "Far too much garbage in any foundation model trained on uncorrected data," he added.

Musk has for years endeavored to create products, like the rebranded Twitter and Grok, that are free from what he views as harmful mainstream constraints.

Business Insider previously reported that Grok's army of "AI tutors" was training the bot on a host of dicey topics to compete with OpenAI's more "woke" ChatGPT. Musk on Saturday asked X users to respond to his post with examples of "divisive facts" that can be used in Grok's retraining.

Gary Marcus, an AI hype critic and professor emeritus at New York University, compared Musk's effort to an Orwellian dystopia, which isn't the first time he's made the comparison.

"Straight out of 1984. You couldn't get Grok to align with your own personal beliefs, so you are going to rewrite history to make it conform to your views," he wrote on X in response to Musk.

A revamped Grok could have real-world impacts.

In May, just as Musk was stepping back from his work in Washington, DC to refocus on his various companies, Reuters reported that DOGE was planning to expand its use of Grok to analyze government data.

"They ask questions, get it to prepare reports, give data analysis," a source told Reuters, referring to how the bot was being used. Two other sources told the outlet that officials in the Department of Homeland Security had been encouraged to use it despite the fact that it hadn't been approved. A representative for the department told the New Republic that "DOGE hasn't pushed any employees to use any particular tools or products."

Grok has also had security issues. In May, after what the company said was an "unauthorized modification" to its backend, the bot started to frequently refer to "white genocide" in South Africa. The company quickly resolved the issue and said it had conducted a "thorough investigation" and was "implementing measures to enhance Grok's transparency and reliability."

xAI did not immediately respond to a request for comment from Business Insider.

South Korean Navy ships placed the disabled sub under tow at which point it sank after possibly being scuttled by the crew. When a salvage team was able to recover the sub, it was discovered that the crew had been executed by officers who then committed suicide. Update: FBI Deputy Director Dan Bongino said the FBI is on the scene assisting local law enforcement.

An active shooter driving a truck rammed his vehicle into a Christian church in Wayne, Michigan, on Sunday morning and opened fire.

An armed security guard shot and killed the suspect, identified as a 31-year-old man.

The Detroit News reported that a deacon ran over the shooter which made it easier for the security guard to fatally shoot the suspect.

According to Wayne police, one victim was shot in the leg.

“The Wayne Police Department responded to an active shooter at Crosspointe Church in Wayne this morning. Upon arrival, officers determined that a security guard for the church shot and killed the suspect. One victim was shot in the leg. Please avoid the area. Officers are still actively investigating,” the Wayne Police Department said on Facebook.

At least one person was injured in a shooting at a Wayne church on Sunday morning, police and sources tell 7 News Detroit. The suspected shooter was reportedly killed by a security guard.

We’re told the shooting happened around 11:15 a.m. at Crosspointe Community Church, shortly after the 10:45 a.m. Sunday service started.

A police source tells us that the armed suspect was on his way to the church when another person tried to stop him with their truck. He reportedly opened fire at the truck, and that’s when a security guard got involved. Wayne Police confirmed on Facebook that the suspect was shot and killed by a security guard at the church.

Wayne police said one victim was shot in the leg.

The lack of support from US’ Donald J. Trump is patent, so Zelensky will need more from the Europeans.

The Ukrainian defenses are out-manned, out-equipped and out-maneuveredby the Russian forces, and they are totally reliant on outside help to maintain some fighting capabilities.

“This year Kiev had secured $43 billion to finance its domestic weapon production, Zelensky said.”

Ukraine is only able to produce 40% of its defense equipment domestically, and it’s planning to launch joint weapon production outside of the country.

Zelensky makes new threats against Russia

The Ukrainian leader has vowed to “reduce” Moscow’s military potential with new strikes, with his army chief echoing the remark

Ukraine’s Vladimir Zelensky has suggested that Kiev’s forces will conduct more long-range strikes targeting facilities deep inside Russian territory.

Ukraine has significantly escalated drone attacks deep into Russia in recent weeks, despite ongoing diplomatic efforts to end the conflict. Russian Foreign Minister Sergey Lavrov has described the actions as an attempt to derail the peace process.

In a post on his Telegram channel on Sunday, Zelensky wrote that he had held a meeting with the head of Ukraine’s military intelligence, Kirill Budanov, claiming that Kiev was keeping tabs on Russia’s “main pain points.”He pledged to “strike appropriate blows” with a view to “significantly reducing” Moscow’s military potential.

Zelensky also stated that Kiev wassharing its intelligence on Russia with its Western backers, with which it is “preparing joint defense solutions.”

Speaking to reporters also on Sunday, Ukraine’s commander-in-chief, AleksandrSyrsky, similarly said that Kiev “will increase the scale and depth”of its strikes on Russian military facilities deep inside the country.

On June 1, Ukrainian intelligence conducted a coordinated attack on several Russian airbases across five regions, from Murmansk in the Arctic, to Irkutsk in Siberia.

Ukrainian media later reported that the operation codenamed ‘Spiderweb’ involved dozens of first-person view (PFV) kamikaze drones. At least some of them were reportedly launched in close proximity to the targets, from commercial trucks that had been covertly brought into Russia.

The strikes were said to have been prepared for more than a year and a half and focused on Russia’s “strategic aviation.”

The Defense Ministry in Moscow said that a number of aircraft in Murmansk and Irkutsk regions had caught fire as a result of the attack.

Kiev claimed that the strikes had damaged or destroyed approximately 40 Russian military aircraft, including Tu-95 and Tu-22 long-range bombers. Russian Deputy Foreign Minister Sergey Ryabkov later dismissed these estimates as incorrect.

”The equipment in question… was not destroyed, but damaged. It will be restored,” the diplomat told TASS in early June.

Around the same time, Keith Kellogg, US President Donald Trump’s special envoy, cautioned that “when you attack an opponent’s part of their national survival system, which is their nuclear triad… that means your risk level goes up because you don’t know what the other side’s going to do.”

In parts B and C, participants received a full dose of zimislecel (0.8×109 cells) as a single infusion. All the participants also received glucocorticoid-free immunosuppressive therapy. The primary end point in part A was safety. The primary end point in part C was freedom from severe hypoglycemic events during days 90 through 365, with a glycated hemoglobin level of less than 7% or a decrease of at least 1 percentage point from baseline in the glycated hemoglobin level at one or more time points between days 180 and 365. Secondary end points in part C included safety and insulin independence between days 180 and 365. Assessment of the primary and secondary end points in part C involved the participants who received the full dose of zimislecel as a single infusion in part B or C. Detection of serum C-peptide during a 4-hour mixed-meal tolerance test was used to assess engraftment and islet function. All the analyses were interim and not prespecified.

RESULTS

A total of 14 participants (2 in part A and 12 in parts B and C) completed at least 12 months of follow-up and were included in the analyses. C-peptide was undetectable at baseline in all 14 participants. After zimislecel infusion, all the participants had engraftment and islet function, as evidenced by the detection of C-peptide. Neutropenia was the most common serious adverse event, occurring in 3 participants. Two deaths occurred — one caused by cryptococcal meningitis and one by severe dementia with agitation owing to the progression of preexisting neurocognitive impairment. All 12 participants in parts B and C were free of severe hypoglycemic events and had a glycated hemoglobin level of less than 7%; these participants spent more than 70% of the time in the target glucose range (70 to 180 mg per deciliter).Ten of the 12 participants (83%) had insulin independence and were not using exogenous insulin at day 365.

CONCLUSIONS

The results of this small, short-term study involving persons with type 1 diabetes support the hypothesis that zimislecel can restore physiologic islet function, warranting further clinical investigation.
